DESCRIPTION OF INTERESTS
Extraintestinal isolates of Escherichia coli Urinary tract infection, pulmonary infection, bacteremia Microbial pathogenesis Vaccine development
Strains of E. coli that cause disease in humans can be broadly divided into two groups. The first group contains the enteric pathogens and the second group consists of those strains that cause extraintestinal infection. The infectious syndromes due to extraintestinal isolates of E. coli range from uncomplicated cystitis to bacteremia with subsequent septic shock and death. The clinical and economic impact of infectious syndromes caused by this cohort of pathogens is enormous, despite the availability of efficacious antimicrobial therapy. I believe that an understanding of the pathogenic process utilized by these organisms will be critical for the logical development of new preventative and/or therapeutic strategies. Our research goal to identify bacterial determinants and to assess: a) in vitro and in vivo their relative importance in the pathogenesis of infection and b) their potential as vaccine candidates. A bacteremic strain is being used as a model pathogen. A genetic, molecular biology approach is beingemployed to achieve this goal. Ongoing projects include: 1) identification of genes required for growth and survival in urine and blood, 2) identification of genes induced in vivo, 3) identification of antigenic determinants expressed in vivo, 4) the role of capsular polysaccharide and lipopolysaccharide, and 5) molecular epidemiologic studies of E. coli strains from women with urinary tract infection. |
